Dapoxetine, the first treatment developed specifically for premature ejaculation, has no significant interaction when taken concurrently with the phosphodiesterase type-5 inhibitors sildenafil citrate (Viagra) and tadalafil (Cialis), according to results presented here yesterday. A drug for premature ejaculation would likely be used in combination with PDE-5 inhibitors by at least some patients. To evaluate the potential for drug interactions and the tolerability of the combinations, investigators randomized 22 healthy men aged 18 to 45 years to one of three treatments: dapoxetine, 60 mg, or dapoxetine, 60 mg in combination with tadalafil, 20 mg, or sildenafil, 100 mg.

For decades, clinical trials with psilocybin, LSD, and DMT have required participants to discontinue antidepressants (SSRIs, SNRIs, MAOIs, etc.) weeks before an entheogen session.
ATC (Anatomical Therapeutic Chemical Classification)
To “avoid serotonin syndrome” and “preserve the full psychedelic experience.” But the latest scoping review by Tap et al. (2025, Journal of Psychopharmacology) suggests this protocol may be both outdated and harmful. After analyzing 19 human studies across psilocybin, LSD, DMT, and ayahuasca, the authors found that: 🧠 Safety: There’s no robust evidence that taking antidepressants with classic entheogens increases the risk of serotonin toxicity — particularly with psilocybin. Most side effects (headache, mild nausea, temporary increases in blood pressure) were comparable to placebo. 💫 Efficacy: Even when antidepressants slightly blunted the intensity of the entheogenic experience, the therapeutic outcomes for depression and anxiety remained significant.

In some studies, patients on antidepressants improved as much or more than those who discontinued them. ⚠️ Risks of discontinuation: Forcing patients to stop antidepressants can lead to relapse, withdrawal symptoms, and suicidal ideation — all before entering a psychologically open and vulnerable state. In other words: the “washout” phase may do more harm than the drug combination itself. 🌱 The takeaway: Continuing antidepressants during entheogen-assisted therapy may actually increase accessibility and safety for millions who currently can’t participate in research or treatment. This isn’t just a pharmacological question — it’s an ethical one. Tadalafil also had no impact on dapoxetine area under the curve. Exposure to dapoxetine, as reflected by AUC, increased by 20% in combination with sildenafil, but Dr.

Abstract: Two simple UV-Spectrophotometric methods have been developed for simultaneous determination of sildenafil citrate and dapoxetine HCl in pharmaceutical formulation. For both the methods stock solutions were prepared in methanol followed by the further required dilutions with methanol. Proposed Vierodt’s method and double point standardization method, the max for the estimation of sildenafil citrate and dapoxetine HCl were selected at 292nm and 231nm respectively. In both methods, the linearity range lies between 2-20 µg/mL for sildenafil citrate and 1.2-12 µg/mL for dapoxetine HCl at their respective wavelengths. By Vierodt’s method the percentage of sildenafil citrate and dapoxetineHCl was found to be 99.25%, dapoxetine HCl 99.08% respectively.
